Retatrutide is a triple agonist — one peptide chain acting at three receptors: GIP, GLP-1 and glucagon. That is a step beyond tirzepatide, which addresses two, and beyond semaglutide, which addresses one.
Each receptor added multiplies the design constraints, since the same sequence must satisfy every binding site simultaneously. It is an investigational compound and has not been approved as a medicine anywhere, which distinguishes it from the other two despite their being discussed together.
