Two hormones, two receptors

The pituitary gland responds to more than one signal. One is GHRH, from the hypothalamus. Another is ghrelin, the hormone the stomach releases when it is empty, which acts at a separate receptor. CJC-1295 is built on the first; Ipamorelin mimics the second. That is the whole reason they are discussed as a pair: they are aimed at different receptors rather than competing for the same one.

Ipamorelin

A five-amino-acid peptide made by Novo Nordisk, the company behind Ozempic, and described in 1998 as “the first selective growth hormone secretagogue”. “Selective” was the point: earlier compounds acting at the ghrelin receptor also raised cortisol and prolactin, and ipamorelin was designed not to. Its one mid-stage trial, in 2008–09 in patients recovering from bowel surgery, did not beat placebo, and development stopped.

CJC-1295

A modified 29-amino-acid fragment of GHRH. It is sold with or without DAC, a chemical hook that lets it bind a blood protein; the difference is large enough to have its own page. Its developer stopped the programme in 2006.
