Origins

When was PT-141 created?

PT-141 is not a fresh design. It is what Melanotan II turns into in the body, isolated and developed as a compound of its own.

The short answer

Date The late 1990s
Who Palatin Technologies
Where New Jersey, United States
Origin Derived from a larger natural molecule, then made synthetically.

Where it came from

When Melanotan II is broken down, one of the things it becomes is the deamidated form — the amide group at one end replaced by a plain acid. That metabolite turned out to be active in its own right, and rather more interesting for one particular purpose than the parent compound.

Palatin Technologies took it forward under the code PT-141. The line of descent is worth stating plainly because it is usually blurred: alpha-MSH is the natural hormone, Melanotan II is the cyclic analogue built from it at Arizona, and PT-141 is a breakdown product of Melanotan II. Three compounds, one lineage, and the two synthetic ones differ by a single functional group.

The late 1990s is the period the development work began; the compound had a long path through trials afterwards, including a change of route of administration partway.

Why it is called that

PT for Palatin Technologies, 141 for the compound number. The international nonproprietary name is bremelanotide, and the approved product is sold as Vyleesi.

Where it stands now

Bremelanotide was approved by the FDA in 2019. It is not approved in New Zealand. Material supplied for laboratory work is not the approved product.

What this page does not say

Nothing above is a statement about what PT-141 does in a body, and none of it is guidance on use. A compound’s history is a record of laboratories and dates; it is not evidence for anything, and the fact that a programme was looking for something is not a finding that it was found.
