When was thymosin alpha-1 discovered?

Thymosin alpha-1 was pulled out of a crude thymus extract called thymosin fraction 5, after years of separating it into pieces.

Where it came from

Goldstein’s group had been working through calf thymus extract since the mid-1960s, separating it into progressively cleaner fractions. Fraction 5 was the one that held the activity they were following, and it was not one substance: it contained a whole set of peptides, which were named alphabetically by where they landed on an isoelectric focusing gel.

Thymosin alpha-1 is the acidic one that came out of that separation in 1972 — a twenty-eight-residue chain with an acetylated N-terminus. The naming convention is the reason for a persistent confusion in this field: thymosin alpha-1 and thymosin beta-4 share a name because they came out of the same tube, not because they are related. They are products of entirely different genes.

That distinction is why this site keeps thymosin alpha-1 and TB-500 on separate pages rather than treating them as two members of one family.

Why it is called that

Thymosin after the thymus. Alpha for where it focused on the gel, 1 for the first of the alpha group. The international nonproprietary name is thymalfasin.

Where it stands now

Thymalfasin is an approved medicine in a number of countries, though not in New Zealand. Material supplied for laboratory work is not that product.
